摘要
选取4只安装有永久性瘤胃瘘管的中国美利奴羊,采用4×4拉丁方设计,在绵羊日粮中分别添加10g/d,20g/d,40g/d的大豆磷脂,以空白为对照,采用PCR-DGGE技术研究不同水平的大豆磷脂对瘤胃微生物多样性的影响。结果显示:在绵羊日粮中添加大豆磷脂后,瘤胃细菌、真菌和原虫DGGE图谱中电泳条带数和迁移率均存在一定差异;微生物丰度分别为35~46、23~38和16~21条/泳道,香浓-威纳多样性指数分别为3.64~3.67、3.28~3.36和2.85~2.92。与对照组相比,添加大豆磷脂各组的瘤胃细菌丰度有不同程度下降,瘤胃真菌和原虫丰度均有不同程度升高,尤以20 g/d组变化较为明显。
Four Chinese Merino male sheep fitted with rumen fistula were used in a 4 x 4 latin square designed experiment tostudy the effects of soybean lecithin on rumen microbial diversity. The four treatments were the basal diet (the control) and the basal diet supplemented with 10g/d, 20g/d,4Og/d soybean lecithin. The ruminal microbial community was analyzed by polymerase chainreaction denaturing gradient gel electrophoresis (PCR-DGGE) Results showed that on the number and migration rate of DNA bands on the DGGE of rumen bacteria, fungi and protozoa changed somewhat. DNA bands and Shannon-Wiener index of lanes of bacteria, fungi and protozoa were 35-46,23-38 and 16-21 ; 3.64-3.67,3.28-3.36 and 2.85 ~2.92, respectively. Compared with the control group, the abundances of rumen bacteria decreased,while that of fungi and protozoa increased by soybean lecithin supplementation, especially in the 20 g/d group.
