摘要
目的建立一种简便、快速、准确检测人心肌肌钙蛋白 T(c Tn T)的胶体金免疫层析法 (GICA)。方法制备的胶体金标记抗 c Tn T单克隆抗体 3F7,生物素标记另一株抗体 2 H8,链霉亲和素结合于硝酸纤维素膜上 ,制成免疫层析试纸条。血清中 c Tn T与测试条两种抗体结合后 ,沿硝酸纤维素膜移动 ,与链霉亲和素交联形成肉眼可见的红色线条。结果测试条灵敏度可达 0 .5 ng/ml。检测 30例急性心肌梗塞 (AMI)患者血清 c Tn T水平 ,并与国外同类产品比较 ,符合率达 92 %。结论本方法特异性强、灵敏度高、简便快速 。
ObjectiveTo develope a sensitive,rapid and simple gold immunochromatography assay(GICA)for determination of human cardiac troponin T (cTnT)in sera.MethodsColloidal gold was prepared and coupled with anti cTnT monoclonal antibody 3F7.2H8,another monoclonal antibody,was biotinylated,streptavidin solidified to nitrocellose to prepare a test strip.cTnT in sera which combined with 3F7 and 2H8,migrates up the nitrocellulose strip after combination of serum cTnT and the two antibodies fixed on the test strip,two red lines were detectable to naked eyes.This was taken as positive.ResultsThe sensitivity of GICA was 0 5 ng/ml.The Level of cTnT in 30 patients with acute myocardial infarction(AMI)were measured by this assay and also by using test kit supplied by Boehringer Mannheim Corp.The corresponding rate between the results using this method and that obtained by using products supplied by Boehringer Mannheim Corp was 92%.ConclusionThis is asensitive,specfic,simple and rapid assay for measuring cTnT.It may be used for early diagnosis of AMI.
