摘要
目的 构建人心肌肌钙蛋白T(cTnT)亚型的重组载体。 方法 应用RT-PCR技术,从人胚胎心肌组织中扩增cTnT1及cTnT2基因,连接到T Easy vector载体上,筛选重组子。结果 成功地扩增出cTnT1(764 bp)及cTnT2(124)基因片段,经测序证实,构建pExSecI-cTnT1表达载体并获得表达。结论 获得单一亚型的cTnT1及cTnT2基因片段,为研究心力衰竭时的胚胎型基因重构与发病关系提供了依据。
Objective To construct recombinant vector for human fetal cardiac troponin T (cTnT) isoform. Methods The cTnT1 and cTnT2 genes were amplified from human fetal cardiac muscular tissue by RT-PCR, and then cloned into T Easy vector. Results The cTnT1 (764 bp) and cTnT2 (124 bp) genes were successfully amplified and their sequences tested, which were cloned into pExSecI vector and expressed by induction at IPTG for 4 h. Conclusions cTnT1 was highly expressed in BL21 transformant with expression level above 18%. These results prepared molecular basis for further research of human cardiac troponin T isoform expressed during heart failure.
