红花多糖对人胃癌SGC-7901细胞线粒体膜电位及增殖的影响 被引量:10

摘要 目的体外观察红花多糖(SPS)对人胃癌SGC-7901细胞线粒体膜电位(MMP)及细胞增殖的影响,初步探讨SPS对SGC-7901细胞的抑制机制。方法不同浓度的SPS处理SGC-7901细胞,倒置显微镜观察SGC-7901细胞的形态学变化,透射电镜观察SGC-7901细胞超微形态结构变化,罗丹明123染色观察SGC-7901细胞MMP变化,MTT比色法检测SGC-7901细胞的增殖。结果倒置显微镜观察到经SPS处理的贴壁细胞体积变小,部分细胞崩解,有凋亡小体形成,随剂量增加愈加明显;透射电镜观察到经SPS处理的细胞线粒体肿胀、胞核浓缩偏移,呈凋亡形态改变;罗丹明123染色可见经SPS处理的细胞内荧光强度显著下降,该作用呈明显的时间依赖性;MTT检测显示经SPS处理的细胞体外增殖受抑制,该作用呈明显的剂量依赖性。结论 SPS可以降低人胃癌SGC-7901细胞MMP,同时能明显抑制SGC-7901细胞体外增殖并可诱导细胞凋亡,该作用呈一定的时间与剂量依赖。 Objective To observe the effects of Safflower polysaccharide (SPS) on the mitochondrial membrane potential (MMP) and proliferation of gastric cancer SGC -7901 cells; thus to investigate the inhibitory effects of SPS on gastric carcinoma cell line SGC -7901. Methods SGC -7901 cells were processed with different concentrations of SPS. The morphology and ultrastructure changes of SGC -7901 cells were observed with inverted microscope and transmission electron microscopy, respectively; as the MMP and cell proliferation rate were assessed with Rhodamine 123 assay and MTY assay, respectively. Results Typical morphological changes of apoptosis were observed, including reduction in volume and presence of apoptotic bodies; furthermore, mitochondria swelling, nuclear enrichment, etc. were also observed. The fluorescence expression of MMP was markedly reduced in Rhodamine 123 assay. Significant inhibition on cell proliferation of SGC - 7901 cells by SPS was observed in time - and dose - dependent manners. Conclusion SPS inhibits the MMP of gastric carcinoma cell line SGC - 7901, also reduces proliferation and induces apoptosis in vitro in time - and dose - dependent manners.
