摘要
目的基于Ras同源基因(Rho)/Rho激酶信号通路对放射性心肌纤维化发病机制及当归红芪多糖的干预作用进行探索。方法首先建立放射剂量梯度,通过筛选放射性心肌损伤的最佳浓度,建立放射性心肌损伤模型。通过法舒地尔、不同浓度当归红芪多糖溶液进行干预,同时采用Western Blot、QT-PCR、流式细胞术3种实验手段检测Rho/Rock及其下游通路的活化、心肌细胞周期、心肌纤维化的改变。结果当归红芪多糖干预后Rho/Rock及其下游通路的活化明显抑制,细胞周期阻滞明减轻,心肌纤维化被逆转。结论当归红芪多糖可能通过抑制Rho/Rock及其下游通路的活化、减轻细胞周期阻滞等逆转放射性心肌纤维化。
Objective Based on Rho/ROCK pathway,to explore the pathogenesis of radiation-induced myocardial fibrosis and the intervention of Angelica Hedysari polysaccharide.Methods The radiation dose gradient was established so as to select the best concentration and establish the radiation myocardial injury model.Western blot,QT-PCR and flow cytometry were used to detect the activation of Rho/rock and its downstream pathway,myocardial cell cycle and myocardial fibrosis.Results The activation of Rho/rock and its downstream pathway were significantly inhibited,cell cycle arrest was reduced and myocardial fibrosis was reversed.Conclusion Angelica Hedysarum polysaccharide may reverse the radiation myocardial fibrosis by inhibiting the activation of Rho/rock and its downstream pathway,reducing cell cycle arrest and so on.
