川芎嗪和参麦注射液对老龄大鼠脑缺血再灌注多器官损伤的作用 被引量:39

Effect of ligustrazine and Shenmai injection(参麦注射液) on multiorgans injury induced by cerebral ischemiareperfusion in senile rats

摘要 目的 :探讨老龄大鼠脑缺血再灌注所致多器官损伤机制 ,观察川芎嗪和参麦注射液及其配伍对多器官损伤的保护作用。方法 :老龄 (2 0月龄以上 )大鼠分为模型组、对照组、尼莫通组、川芎嗪组、参麦组和川芎参麦组 (川参组 ) ,检测各组药物对老龄大鼠全脑缺血 30分钟及再灌注 6 0分钟后多巴胺 (DA)、去甲肾上腺素(NE)、皮质醇 (Co)、一氧化氮 (NO)和肿瘤坏死因子 (TNF)水平 ,同时观察脑、心、肝、肺、肾、胃、小肠病理改变。结果 :模型组大鼠脑组织均出现病理改变 ,交感肾上腺系统兴奋增强 ,脑组织中 TNF含量增高 ,血清中 NO含量降低。用药各组老龄大鼠脑组织损伤均有明显减轻 ,下丘脑 NE和脑组织 NO含量增高及下丘脑 DA水平降低 ,川芎嗪组、参麦组及川参组下丘脑 NE含量和 DA水平接近对照组 ,参麦组血中 Co水平和川参组脑组织TNF含量降低 ,川芎嗪组血清中 NO水平增高。模型组大鼠多个器官较对照组出现病理改变 ,交感肾上腺系统兴奋增强 ,各组药物对器官损伤均有不同程度的保护作用和对交感肾上腺系统有明显的调节效果。结论 :老龄大鼠脑缺血再灌注损伤与交感肾上腺系统兴奋增强和 NO含量降低及 TNF增高有关 ,川芎嗪和参麦注射液及其配伍通过调节交感肾上腺系统和提高 NO水平以防护老龄大鼠脑缺血再灌注脑组? Objective:To study the effect of Ligustrazine(LT),Shenmai injection(SM,参麦注射液) and the integration of ligustrazine and Shenmai injection (LS) on multiorgans injury induced by cerebral ischemiareperfusion in old rats,in order to offer the evidences for traditional Chinese medicine and Chinese herbs to prevent and treat cerebrovascular disease and secondary multiorgans injury in the elderly.Methods:Aged rats (20 months old or more) were divided into six groups:model group,control group,LT group,SM group,LS group and Nimotop group.The effect of the drug in each group on the pathologic alterations of brain,heart,liver,lung,kidney,stomach and small intestine and the changes in the concentration of dopamine(DA),noradrenalin(NE),cortisol(Co),nitric oxide(NO) and tumor necrosis factor(TNF) in blood and hypothalamic after fully cerebral ischemia for 30 minutes and reperfusion for 60 minutes in aged rats were observed.Results:Pathological change was found in the rats′brain tissue in the aged model,the excitability of sympatheticadrenal system was enhanced,brain tissue TNF content increased,the serum NO level was lower.The brain injury was improved in all treated group,the hypothalamus NE and brain tissue NO content was higher,the hypothalamus DA level was lower,the NE and DA levels in the LT group,SM group and the LS group were close to those in the aged control group.The Co content in the SM group and the brain tissue TNF in LS group was decreased,the level in serum NO in LT group was higher.Several organ injuries in model group were more obvious and excitation of sympathetic nervesuprarenal gland system were more excessive than those in control group.The drug in each group had in preventing effects in different degree on injuried organs and had the adjustment effects on sympathetic nervesuprarenal gland system significantly.Conclusions:The brain ischemiareperfusion injury is correlated with the enhanced excitability of sympatheticadrenal system,NO level decrease and TNF content increase.LT,SM and LS can prevent this injury in aged rats via adjusting the excitability of sympatheticadrenal system and increased NO level,the effect of SM on the stress reaction of hypothalamuspituitaryadrenocortical axis and LT on TNF was obvious.The mechanisms that LT,SM and LS have the preventing effects on multiorgans injuries induced by cerebral ischemiareperfusion in aged rats are related to effects of suppressing the excessive excitation of the sympathetic nervesuprarenal gland system.
