内生菌液体发酵提高穿山龙提取物中薯蓣皂苷元的含量 被引量:3

Increasing Diosgenin Content in Extract of Dioscorea Nipponica by Liquid Fermentation of Endophyte

摘要 目的:分离提取穿龙薯蓣内生真菌,筛选能够发酵提高穿山龙药材提取物中薯蓣皂苷元含量的功能菌种。方法:采用PDA培养基分离纯化穿龙薯蓣内生真菌,将内生真菌与灭菌后穿山龙药材提取物在恒温振荡培养箱中,120 r/min,28℃培养7天。以高效液相法测定发酵物中薯蓣皂苷元的含量,并与原药材提取物、灭菌药材提取物以及未灭菌药材提取物的发酵物进行比较。结果:分离提取穿龙薯蓣内生真菌102种,其中C39菌能够稳定提高灭菌后穿山龙提取物中薯蓣皂苷元的含量,提高幅度达原药材提取物的20%-30%。结论:经液体发酵,穿龙薯蓣内生真菌C39能够稳定提高穿山龙药材提取物中薯蓣皂苷元的含量。 Objective: To isolate and screen the functional endophyte from Dioscorea nipponica for increasing the diosgenin content in the extract of its rhizome by fermentation. Methods: The endophytes were isolated from Dioscorea nipponica with PDA culture medium. The extract of Dioscorea nipponica was sterilized by moist heat at 121 ℃, then fermented with the endophytes in a shaking incubator at 120 r/min, with constant temperature 28℃ for seven days. The diosgenin contents of the fermentation products were analyzed with HPLC and further compared with that of the original rhizome extract, the sterilized rhizome extract and the ferment of unsterilized extract. Results: 102 endophytes were isolated from Dioscorea nipponica. Among them, endophyte C39 showed the property that stably increased the diosgenin content in sterilized extract of Dioscorea nipponica to 20%-30% compared with the original rhizome extract. Conclusions: Endophyte C39 increase the diosgenin content in the extract ofDioscorea nipponica bv liouid fermentation.
