摘要
目的 从分子水平认识转化生长因子 β1(TGFβ1)在骨折愈合过程中的作用。方法 采用地高辛 (DIG)标记寡核苷酸探针的原位杂交方法 ,光镜观察成年新西兰白兔桡骨中段骨折愈合过程中 ,TGFβ1、mRNA在骨痂区各组织细胞中的表达变化情况。结果 在骨折愈合早期 ,可见少量TGFβ1表达阳性的单核细胞、间充质细胞和血管内皮细胞。在膜内成骨期 ,TGFβ1在成骨细胞内有显著表达 ,尤其是多角形丰满成骨细胞。在成软骨期 ,较成熟软骨细胞内有较高表达。在软骨内成骨期 ,成骨细胞内有高表达 ,而在肥大的软骨细胞内几乎无表达。结论 TGFβ1在整个骨折愈合过程中的每个阶段都具有重要作用 ,影响着细胞分化、增殖与成熟以及基质的形成。
Objective [HT5”SS]To investigate the role of transforming growth factor β 1(TGF β 1)in fracture healing on the molecular level.[HT5”H]Methods [HT5”SS]The local expressions of TGF β 1 in callus during the process of healing of adult New Zealand white rabbits were investigated through optical microscope,using in situ hybridization of DIG labeled oligonucleotide probe.[HT5”H]Results [HT5”SS]At the early stage of fracture healing,TGF β 1、mRNA were presented in a few monocytes,mesenchymal cells,and endothelial cells.At the stage of intramembranous bone formation,TGF β 1 expressed notably in the osteoblasts,especially in the multiangle plentiful osteoblasts.At the stage of chondrogenesis,nearly mature chondrocytes were strongly positive.While at the stage of endochondral ossification,the hypertrophic chondrocytes were almost negative.TGF β 1 expressed strongly in osteoblast at this stage.[HT5”H]Conclusion [HT5”SS]TGF β 1 acted as an important role in every stage of fracture healing by affecting the differentiation,proliferation,maturation of cells and the formation of matrix.
