摘要
目的:观察瘦素对乳腺癌细胞(T47D)Survivin表达的影响。方法:取对数生长期人乳腺癌细胞T47D,以5×104个/孔种植于48孔板,分别进行以下处理,C组:为对照组,饥饿后不做任何处理;L组:以10nmol/L瘦素处理细胞。蛋白质印迹法测定Survivin蛋白表达的水平。RT-PCR测定Survivin mRNA的表达。结果:蛋白质印迹法检测结果示,T47D细胞经10nmol/L瘦素作用2h后,Survivin蛋白的表达增强。采用瘦素(10nmol/L)分别作用T47D细胞30min和60min后,瘦素能促进T47D细胞Survivin mRNA的表达,与对照组相比Survivin mRNA的表达量是对照组的(2.15±0.19)和(4.55±0.23)倍。结论:瘦素可以上调乳腺癌T47D细胞Survivin的表达。
OBJECTIVE: To detect the effect of lepin on Survivin expression in breast cancer cells (T47D). METH- ODS: To collect breast cancer cells T47D in the logarithmic growth phase,and culture them at 5 )〈 104/ well in 48-well plates,respectively, in the following processing. C group: the control group, without any treatment after starvation; L group:cells treated with 10 nmol/L Leptin. The Survivin protein expression was quantified by Western-blot assay. In addition, the Survivin mRNA expression was detected by real-time quantitative reverse transcription polymerase chain reaction (RT-PCR). RESULTS: Western blot analysis showed that Survivin protein expression in T47D cells was upregulated afer treatment by 10 nmol/L Leptin for 2 h. RT-PCR showed that Leptin (10 nmol/L) could promote the expression of Sur vivin mRNA in T47D cells after treatment for 30 min and 60 min, compared with the control group. After treatment by Leptin (10 nMol / L) for 30 rain and 60 min,Survivin mRNA expression was 2.15±0.19 and 4.55±0.23 times higher than those of the control group. CONCLUSION: Leptin increases the expression of Survivin in T47D breast cancer cells.
