瘦素与前列腺癌激素依赖性关系的研究 被引量:2

摘要 目的观察瘦素对前列腺癌细胞增殖的促进作用,探讨其与前列腺癌激素依赖性的关系。方法采用四甲基偶氮唑蓝(MTT)法检测瘦素作用不同时间后雄激素依赖性前列腺癌细胞株22RV1和雄激素非依赖性前列腺癌细胞株PC-3的增殖率;用MTT法检测不同浓度瘦素及双氢睾酮(DTH)作用后前列腺癌细胞株22RV1和PC-3的增殖率。结果瘦素作用于PC-3细胞不同时间后吸光度(OD值)均明显增高(P<0.05),在48 h达到顶点;且随瘦素浓度逐渐增高,其促进PC-3细胞增殖率亦逐渐增高;而瘦素对22RV1细胞作用不同时间后OD值增高不明显(P>0.05)。DTH对22RV1细胞增殖作用最明显,增殖率随其浓度增高而逐渐升高,但其促进PC-3细胞增殖率明显较低(P<0.01)。结论瘦素对雄激素非依赖性前列腺癌细胞增殖具有明显的选择性促进作用,其可能与前列腺癌雄激素非依赖性的发生发展有密切关系。 [ Objective ] To observe the proliferation promotive effect of leptin to prostate cancer cell. [ Methods ] MTY assay was used to examine the proliferation rate of prostate cancer cells growth after treatment with leptin for different long time. MTY assay was used to examine the proliferation rate of hormonal dependent prostate cancer cells 22RV1 and hormonal independent prostate cancer cells PC-3 growth after treatment with leptin or dihy- drotestosterone. [ Results ] Optical density (OD) increased obviously (P 〈0.05) after PC-3 cell were treated with lep- tin. The value reach to the limit after 48 h. Furthermore, exposure to leptin caused a significant dose-dependent in- crease. Optical density (OD) did not increased obviously (P 〉0.05) after 22RV1 cell were treated with leptin. The proliferation rate of PC-3 cells is lower than (P 〈0.01) 22RV1 cells that is highest after treatment with dihy- drotestosterone. [ Conclusion ] Leptin have apparent selectivity promotive effect to proliferation of hormonal inde- pendent prostate cancer cells, it may has closely correlation with development and progress of hormonal independent prostate cancer.
