摘要
目的观察瘦素对人肺腺癌A549细胞增殖及p-ERK1/2、VEGF表达的影响。初步探讨瘦素在促进肿瘤血管生成中的机制。方法对人肺腺癌A549细胞采用细胞培养,MTT法检测不同浓度瘦素对人肺腺癌A549细胞增殖影响,免疫组织化学法检测VEGF表达、Westernblot方法检测瘦素对p-ERK1/2、VEGF表达的影响。结果在一定范围内,瘦素呈时间、剂量依赖性促进A549细胞的生长(P<0.05);与阴性对照组相比,不同浓度瘦素作用48h后p-ERK1/2、VEGF蛋白表达明显增加并具有剂量依赖性(P<0.05);p-ERK1/2和VEGF之间呈正相关性(r=0.694,P<0.05)。结论在体外瘦素对人肺腺癌A549细胞有明显增殖作用,通过促进p-ERK1/2和VEGF的表达,瘦素在促进肺癌血管的生长机制中可能具有一定作用。
Objective To explore the effect and significance of leptin on the expression of p-ERK1/2 and VEGF in A549 cells.Methods A549 cell line was treated with different concentration of letpin for 24,48 or 72 hours,respectively,and then cellular proliferation was evaluated by MTT method.VEGF expression in tumor cells was detected by immuno-histochemical method,Western blot analysis was used to detect both p-ERK1/2 and VEGF expressions.Results MTT assay showed that Leptin stimulated the proliferation of A549 cells in a does and time-dependent manner in vitro(P〈0.05).The expression of VEGF was upregulated significantly in A549 cells treated by 80ng/ml or 160ng/ml Leptin(P〈0.05).Western blot showed that the protein expression of VEGF was significantly increased in a Leptin dose-dependent way.There was a positive correlation between p-ERK1/2and VEGF(r=0.694,P〈0.05).Conclusion Leptin stimulated proliferation of A549 cells and up-regulated VEGF protein expression partly through ERK1/2 pathway.
