提取-共沸精馏耦合工艺提取挥发油制备颈复康颗粒的抗炎镇痛作用 被引量:3

Anti-inflammatory and Analgesic Effects of Jingfukang Granule by Water Extraction Coupling Rectification

摘要 目的:研究提取-共沸精馏耦合工艺(WER)提取挥发油制备颈复康颗粒的抗炎镇痛作用,初步探讨其作用机制。方法:动物随机分为颈复康颗粒高、低剂量组(12,6 g.kg-1)、WER工艺颈复康颗粒高、低剂量组(12,6 g.kg-1)及空白对照组,各组连续ig给药7 d。采用小鼠耳廓肿胀法、腹腔液渗出法和大鼠足肿胀法研究WER工艺制备的颈复康颗粒的抗炎作用;采用小鼠扭体法、热板法及大鼠温浴法研究其镇痛作用;通过测定大鼠肿胀足白介素1(IL-1)、肿瘤坏死因子(TNF-α)、前列腺素E2(PGE2)含量,分析WER工艺与传统工艺制备的颈复康颗粒对上述各项指标的影响。结果:WER工艺制备的颈复康颗粒高、低2个剂量组均可明显减轻小鼠耳肿胀和大鼠足肿胀(P<0.01,P<0.001),减少小鼠腹腔液的渗出(P<0.05,P<0.01),提高小鼠扭体法、热板法及大鼠温浴法所引起疼痛的痛阈值(P<0.01,P<0.001),降低大鼠肿胀足中IL-1,TNF-α和PGE2含量(P<0.01,P<0.001),且与传统工艺颈复康颗粒同剂量组比较差异有显著性(P<0.05,P<0.01)。结论:WER工艺制备的颈复康颗粒具有更显著的抗炎镇痛作用,机制可能与降低IL-1,TNF-α和PGE2含量有关。 Objective:To study the anti-inflammatory and analgesic effects of Jingfukang Granule by water extraction coupling rectification(WER). Method: The anti-inflammatory effect was evaluated by ear edema induced by dimethylbenzene, celiac capillary vessel permeability induced by acetic acid in mice and foot swelling induced by carrageenan in rats. The analgesic effect was evaluated by writhing method in mice,hot-plate method in mice and water bath method in rats. Result: Compared to Jingfukang Granule, Jingfukang Granule by WER could significantly inhibit the ear edema formation (P<0.01,P<0.001), acetic acid-induced increase in capillary permeability and foot swelling (P<0.05,P<0.01). Jingfukang Granule by WER could also significantly increase the threshold of pain (P<0.01,P<0.001). It could also decrease the contents of interleakin(IL-1),fumor mecrosis factor-α(TNF-α) and prostaglaodin E2(PGE2)(P<0.01,P<0.001). Conclusion : Jingfukang Granule by WER has more significant anti-inflammatory and analgesic effects,which may be related to the inhibition of IL-1, TNF-α and PGE2.
