摘要
目的研究黄芪总苷(TA)对体外培养的人早幼粒细胞白血病细胞株凋亡的线粒体传导通路的影响。方法将不同浓度的TA(200、300、400μg/ml)作用于NB4细胞,体外培养48 h,使用流式细胞仪检测细胞凋亡率及线粒体膜电位的变化;采用RT-PCR测定细胞内Bcl-2和Bax mRNA变化;使用分光光度法测定细胞内caspase-3的活性变化。结果TA作用于NB4细胞48 h后,各浓度组的凋亡率与对照组相比差异均有统计学意义(P<0.05),在TA(200、300、400μg/ml)组凋亡率呈浓度依赖性增加,各组之间差异有统计学意义(P<0.05);与对照组相比,各浓度组Bcl-2 mRNA及线粒体膜电位的表达明显下降,caspase-3的表达明显升高(P<0.05);Bax mRNA的表达无明显差异。结论 TA可以诱导NB4细胞凋亡,具有体外抗白血病效应,并且可能通过线粒体凋亡途径诱导细胞凋亡。
Objective To study the effects of total astragalosides (TA) on the mitoehondria-dependent apoptotie pathway in NB4 cells. Methods NB4 cells were treated with different concentration (200,300,400 μg/ml) of TA for 48 h . The rates of apoptosis of NB4 cells and the expression of mitoehondrial membrane potential were detected by flow eytometry. The mRNA expression of gene Bcl-2 and Bax were detected by RT-PCR method. The relative activity of easpase-3 was examined by ehromatometry assay. Results After incubation for 48 h, there was significant difference in apoptosis between TA group and blank control ( P 〈 0. 05 ). Besides, TA induced apoptosis of NB4 cells in a dose-dependent manner. In contrast with blank control, the expression of Bcl-2 mRNA and mitoehondrial membrane potential in TA groups were lower(P 〈 0.05) ;the activity of caspase-3 was higher(P 〈 0. 05 ). But there was no significant difference in the expression of Bax mRNA between TA group and blank control. Conclusion TA can induce apoptosis of NB4 cells and may have antileukemia effect in vitro, and TA may induce apoptosis of NIM cells by mitochondria-dependent pathway.
