摘要
目的考察香附挥发油经透皮透过量及其对吲哚美辛体外经大鼠皮肤促渗透作用,为香附挥发油和吲哚美辛经皮渗透制剂的研究提供依据。方法建立吲哚美辛和香附挥发油中主要成分α-香附酮的HPLC法。用改良的Franz扩散池考察α-香附酮和吲哚美辛的体外经皮透过量。结果香附挥发油和吲哚美辛的皮肤渗透行为均符合Higuchi方程,累积渗透量Qn与时间t呈良好的线性关系,香附挥发油皮肤渗透量随浓度增大而增大;5%、3%、1%香附挥发油对吲哚美辛的增渗倍数分别为0.943 1、0.621 1、0.195 9,相应浓度氮酮的增渗倍数分别为1.105 0、0.407 8、0.094 8。结论香附挥发油具有良好的皮肤渗透性,与氮酮的促渗效应相似,可以作为吲哚美辛的透皮促进剂。
Objective To study the percutaneous permeability of Cyperus rotundus volatile oil and the enhancing effects of the volatile oil on percutaneous permeability of indomethacin through rats' skin in vitro,and provide the basis for the transdermal preparations of the volatile oil and indomethacin.Methods The method to determine indomethacin and α-cyperone was established by HPLC.Permeation studies in vitro were carried out by using Franz-diffusion cells to investigate the cumulative permeation content of indomethacin and α-cyperone.Results The permeation process of the volatile oil and indomethacin were concordant with Higuchi equation,Qn showed a good linear relationship with t;The enhancement rates of 5%,3% and 1% volatile oil for indomethacin were 0.943 1,0.621 1,0.195 9,respectively.The corresponding concentrations of Azone were 1.105 0,0.407 8,0.094 8,respectively.Conclusion Cyperus rotundus volatile oil has a good skin permeability and similar enhancing effect with Azone,it can be used as transdermal enhancers for indomethacin.
