复合氨基酸中牛磺酸的高效液相色谱分析 被引量:14

摘要 采用国产C1 8反相柱 ,研究了异硫氰酸苯酯 (PITC)柱前衍生高效液相色谱法测定复合氨基酸中牛磺酸的可行性 ,测定了淡水珍珠中牛磺酸的含量 ,结果表明 ,采用甲醇流动相体系 ,在适宜的色谱条件下 ,牛磺酸可获得较好的分离 ,并且在 8~ 4 0 μg/mL浓度范围内与峰面积成良好的线性关系 ,峰面积定量的相对标准偏差为 0 .7% (n =5) ,牛磺酸的PITC衍生物溶液室温存放 3 6h不变质 ,用本法测得淡水珍珠样品中牛磺酸含量为 0 .0 2 %。 By use of octadecyl (C18) column made in China, a RP-HPLC method using phenylisothiolyanate (PITC) as pre-column derivatizing reagent for the determination of taurine in amino acids was studied and the content of taurine in fresh-water pearls was analyzed. The result indicates that under the suitable condition, the taurine derivatized with PITC can be well separated from other phenylthiocarbamyl (PTC) amino acids by utilizing methanol system of flow phase. The range of linear relationship between the peak area and the concentration of taurine is 8~40 μg/mL and the ralative standard deviation of quantitation of peak area is 0.7%( n =5). The solution of taurine derivatized with PITC can keep stability for 36 hours when stored under room temperature. The content of taurine in sample of fresh-water pearls obtained by this method is 0.02%.
