摘要
目的观察肾衰泄浊汤对肾间质纤维化大鼠肾组织OPN、CD44的调节作用。方法采用单侧输尿管梗阻动物模型,将72只SD大鼠随机分为6组。术后第7天、第14天处死,用原位杂交法检测实验大鼠肾组织OPN、CD44的调节作用。结果术后第7天、第14天,中药各组、苯那普利组与模型对照组大鼠肾组织OPN、CD44mRNA表达较假手术组增强(P<0.01),Ⅰ、Ⅲ型胶原、FN生成增多(均P<0.05)。术后第14天,中药各组和苯那普利组大鼠肾组织OPN、CD44mRNA表达较模型组明显减弱(P<0.05),Ⅰ、Ⅲ型胶原、FN生成明显减少(P<0.05),且大鼠肾组织病理损伤明显改善,以中药高剂量组最为显著,治疗效果亦较贝那普利组更佳(各项指标对比(P<0.05)。结论肾衰泄浊汤可能是通过抑制大鼠肾组织OPN、CD44mRNA的表达,抑制Ⅰ、Ⅲ型胶原、FN等ECM的过度沉积,从而减轻肾间质纤维化。
Objective To observe the effect of Shenshuai Xiezhuo Tang on the expression of OPN/CD44 in renal interstitial fibrosis rats, clarify such party resistance to renal function mechanism of interstitial fibrosis (RIF). Methods Using the Pulitzer group unilateral ureter obstruction (UUO) animal models,72 rats were randomLy divided into six groups.Respectively on postoperative 7 days,the first 14 days of death, the effect of OPN/CD44 in experimental rats kidney tissues by hybridization in situ method were examined. Results For the first seven days after surgery,and the first 14 days,TCM groups,henazepril group,model group rats,the expression of OPN/CD44mRNA in kidney tissues was higher than sham operation group (P 〈 0.01),collagen I and m type, FN formation were increased (P 〈 0.05). And postoperative day 14,TCM groups,benazepril group rats the expression of OPN/CD44mRNA in kidney!tissues were significantly lower than model group (P 〈 0.05),collagen I and m type, FN formation were decreased(P 〈 0.05).And rat renal tissue pathology improved markedly,especially the Chinese medicine high dose group most evidently, and the treatment effect better than benazepril group (each index all contrast, P 〈 0.05). Conclusion Shenshuai Xiezhuo Tang has the ability of inhibiting the expression of OPN/CD44mRNA in kidney tissues,inhibiting collagen I and Ⅲ type,FN etc.exeessive deposition, reduce renal interstitial fibrosis.
