摘要
目的:了解黄芪(AM)对人肾小球系膜细胞(HMCs)分泌基质和表达CD44mRNA的影响。方法:应用不同浓度 AM或 AM血清处理经 PDGF-BB刺激的 HMCs,分别采用 MTT法和 ELISA方法检测 HMCs增殖和Ⅳ型胶原(Col-Ⅳ)分泌,用RT-PC R法检测AM对HMCs表面CD4 4mRNA表达的影响。结果:经AM或AM血清处理后,由PDGF-BB诱导的HMCs增殖与未处理对照组相比呈显著抑制,并呈剂量依赖效应;HMCs分泌Col-AⅣ和CD44mRNA表达水平较未处理对照组相比也有显著下降,并与剂量呈一定相关。结论;AM可抑制HMCs增殖和Ⅳ型胶原蛋白的过度生成,并且这种作用可能部分是通过降低 HMCs表面 CD44 mRNA的过表达而介导的。本研究结果提示,AM对延缓慢性肾脏疾病向肾硬化进展可能有益。
Objective: To investigate the effect of AM on matrix secretion and CD44 mRNA expreSSion in human renal mesangial Pells (HMCh). Methods: Cultured HMCs were treated with AM and AM serum in different concentrations in the presence or absence of PDGF- BB, the proliferation and type Ⅳ collagen secretion (Col - Ⅳ ) in HMCs were measured by MTT assay and ELISA, and expression of CD44 mRNA was estimated by RT - PCR method, respectively. Results: After AM or AM serum treatment, the proliferation of HMCs induced by PDGF - BB was inhibited significantly in a dine - dependent manner compared with untreated control; Col -Ⅳ secretion of HMCs and CD44 mRNA level were significantly decreased in a dose - dependent manner compared with untreated control. Conclusions: AM could inhibit proliferation and matrix over - synthesis of HMCs. And these might be mediated partly by decrease of CD44 mRNA over- expression on the sufface of HMCs. The present report showed that AM could have a beneficial effect on delay that chronic renal diseases progress to glomerulosclerosis.
