摘要
将210名妊娠3~8周妇女静脉混合血清经DE_(52)、ConA-Sepharose CL-4B和Sephadex G-100 3次柱层析后,获得了hCG(-)、具花抑活性的样本孕G-Ⅱ、G-Ⅲ和G-Ⅳ。对照组为37例非孕、健康妇女混合血清。PAGE分析表明孕G-Ⅳ出现1条与孕G-Ⅲ的主要蛋白带相似的带,还有一条小带。SDS-PAGE显示,孕G-Ⅲ、G-Ⅳ均存在1条分子量为1.9×10~4u的蛋白带,且孕G-Ⅳ另2条带分子量均为1.9×10~4u的整数倍,为3.8×10~4u和5.7×10~4u。并测定了EPF活性物孕G-Ⅳ的等电点。
Early pregnancy factor(EPF) was purified from the pooled sera of 210 pregnant women after 3~8 weeks of gestation. Sera from healthy nonpregnant women were used as control. The samples (GⅡ, GⅢ andG Ⅳ)obtained from pregnant women have EPF activity but no hCG activity. Polyaerylamide gel electrophoresis(PAGE) showed that the major bands of pregnant G-Ⅲ and G-Ⅳ have similar position in tube gels. The results of SDS-PAGE showed that pregnant G-Ⅳ has 3 bands of molecular weight: 57.5, 38.0 and 19.0 KD. The pⅠ of pregnant G-Ⅳ is 6.45 and 8.20.
