摘要
以花结抑制试验检测250名妊娠3~8周妇女静脉血混合血清及其经DEAE纤维素离子交换柱层析、ConA-Sepharose亲和层析和Sephadex-G-100凝胶过滤后所得各阶段提取物的花抑滴度,并检测其hCG和蛋白含量。对照组为37名非孕妇女混合血清。结果证明:混合孕血清经DE—52柱层析后未吸附蛋白部分DE-Ⅰ;ConA-Sepharose亲和层析后,DE-Ⅰ-ConA-Ⅰ、DE-Ⅰ-ConA-Ⅱ和SephadexG-100过滤后的GⅡ、GⅢ、GⅣ均具花抑活性;DE-Ⅰ和DE-Ⅰ-ConA-Ⅱ的hCG检测均为阳性;而DE-Ⅰ-Con-A-Ⅰ和GⅡ、GⅢ、GⅣ的hCG均为阴性。提示GⅡ、GⅢ、GⅣ为具花抑活性而hCG为阴性的EPF提取物。
The immunosuppressive early pregancy factor (EPF) present in the serum of pregnant women(250 persons) between 3~8 weeks of gestation was isolated and purified using modified Mehta method. EPF activity, hCG, and the protein content of the fractions were detected. Sera from women without history of pregnancy were used as control. The results showed: EPF with rosette inhibition activity was in the unbound fraction (DE-Ⅰ) from chromatography of the pregnant sera on DE-52, DE-Ⅰ-ConA-Ⅰ, and DE-Ⅰ-ConA-Ⅱ of affinity chromatography on ConA-Sepharose. GⅡ, GⅢ, and GⅣ of gel filtration on sephadex G-100; hCG was detected in DE-Ⅰ and DE-Ⅰ-ConA-Ⅱ, but not in DE-Ⅰ-ConA-Ⅰ, GⅡ, GⅢ, and GⅣ. It was suggested that GⅡ, GⅢ, and GⅣ were the extract of EPF with rosette inhibition activity but with negative hCG.
