摘要
采用CTAB法提取白酒小曲细菌总DNA,用细菌通用引物扩增16S rDNA,并构建16S rDNA克隆文库。采用限制性内切酶片段长度多态性(RFLP)技术对16S rDNA文库进行分析,获得76个克隆的酶切指纹图谱。选择酶切带型不同的阳性克隆进行测序并且构建进化树。结果表明,小曲主要细菌种类为黄单胞菌属、沙雷氏菌属、乳杆菌属、木崎氏菌属和片球菌属,其中优势细菌是以戊糖片球菌为主的乳酸菌。
The total bacterial DNA of Xiaoqu was extracted using CTAB method. The bacterial 16SrDNA fragments were amplified with universal primers, and a 16S rDNA clone library was constructed. The 16S rDNA fragments were analyzed by restriction fragment length polymorphism (RFLP) method. The fingerprints of 74 clones were obtained. The positive clones producing different restriction endonuclease types were sequenced and the phylogeny tree was constructed. The results showed that the members of Xiaoqu bacteria were Xanthomonas, Serratia, Short lactobacillus, Kozakia and Lactobacillus, among which the dominant bacteria was Pediococcus pentosaceus.
