摘要
以郑州市郊冬小麦农田土壤为研究对象,利用改进的土壤DNA提取方法提取土壤微生物基因组,并采用降落式PCR和DGGE电泳技术对细菌16S rDNA V3区进行扩增和产物分离,分析了小麦4个生育时期3个土层深度的细菌群落变化.结果表明,麦田土壤细菌多样性极高,小麦整个生育期土壤中一直存在数种优势菌群.但各时期都有新的不同的细菌出现,整体表现为随着小麦的生长多样性逐渐增加,收获期减少.表层土壤各生育时期菌群数量变化较大,而深层土壤菌群数量变化较小.
Using improved extraction method of soil DNA and soil from the winter wheat fields in Zhengzhou suburb as experiment subject, we extrated the DNA genome from soil microbials. In this paper, the diversity of bacterial community was studied by amplifying the bacterial 16S rDNA V3 fragment with t0uch-down PCR method, and separating the products by denaturing gradient gel electrophoresis (DGGE). Soil samples were taken at three different depths at four different stages. The result showed that, the diversity of soil bacteria is higher in wheat fields. With the growth of wheat, the diversity is increasing gradually, and decreasing in harvest period. There are a few stable and dominant bacteria communitiy in the whole wheat growth period, bat some new different bacteria appeare at every period. The number of bacteria communitiy in top soil changes more greatly than in deep soil in different period.
