摘要
目的:通过观察参地颗粒对抗肾小球基底膜肾炎大鼠血、尿生化指标、肾脏组织病理以及肝细胞生长因子(HGF)和转化生长因子-β1(TGF-β1)在肾组织表达的影响,探讨其对大鼠抗肾小球基底膜肾炎的干预作用。方法:建立大鼠抗肾小球基底膜肾炎模型,随机分为病理对照组、参地颗粒组、肾炎舒组及正常对照组。监测实验第4天及第4、8周24h尿蛋白定量、血生化指标动态变化,观察肾组织病理形态学变化,应用免疫组化法观察大鼠肾组织HGF、TGF-β1的表达。结果:造模后第4天开始,病理对照组、肾炎舒组、参地颗粒组的24h尿蛋白定量、血生化指标较正常对照组比较有显著升高(P<0.05),之后一直维持较高水平;治疗8周后,参地颗粒组的24h尿蛋白定量、血肌酐和尿素氮水平与同期病理对照组、肾炎舒组比较,差异均有显著性意义(P<0.05)。免疫组化显示,病理对照组大鼠的HGF、TGF-β1蛋白表达半定量积分与正常对照组相比有显著性上升(P<0.05);而参地颗粒组和肾炎舒组的HGF半定量积分高于病理对照组(P<0.05),TGF-β1半定量积分低于病理对照组(P<0.05)。结论:参地颗粒对抗肾小球基底膜肾炎大鼠具有较好的肾脏保护作用。其作用机制可能是通过促进肾脏HGF分泌,抑制TGF-β1产生;同时也进一步增强HGF对TGF-β1的抑制作用来发挥对肾脏的保护作用。
Objective: To investigate the influence of Shendi Granules (SG) on expressions of hepatocyte growth factor(HGF)and transforming growth factor-β1 (TGF-β1)as well as blood and urine biochemical indicators in rats with anti-glomerular basement membrane nephritis(anti-GBM nephritis), and to explore the intervention effect on anti-GBM nephritis. Methods: The rats model of anti-GBM nephritis was established, and then were randomly divided into 4 groups, and they were model group, SG group, Shenyan Shu group and normal group. Urinary protein and blood parameters were detected after 4 days, 4 weeks, 8 weeks respectively. The renal pathological changes were observed after HE, PAS and Masson staining. Immunohistochemical assay was adopted to observe HGF and TGF-β1 protein expression in the renal tissue. Results: From day 4 after the modeling, the 24-hour urinary protein (24hUP), serum creatinine(SCr)and blood urea nitrogen(BUN)were significantly higher in model group, SG group and Shenyan Shu group than those in normal group(P〈 0.05), and remained high level from then on. After treatment for 8 weeks, 24hUP, SCr and BUN levels were lower in SG group than those in the model group and Shenyan Shu group (P 〈0.05). The results of immunohistochemical assay showed that the model group had higher semi-quantitation scores of HGF and TGF-β1 protein expression than the normal group(P 〈 0.05), and SG group and Shenyan Shu group had higher HGF expression level while lower TGF-β1 than the model group (P 〈 0.05). Conclusion.. Shendi Granules shows certain renal protective effect on anti-GBM nephritis rats. The mechanism may be relate to elevating the expression of HGF and reducing the production of TGF-β1, simultaneously, Shendi Granules promotes the inhibitive effect of HGF on TGF-β1.
