摘要
目的 探讨内皮素抗血清对低氧缺血脑损伤新生鼠可能的保护作用。方法 应用放射免疫法、硫代巴比妥法及氨基酸自动分析法观察 :(1)正常鼠 (n=7)及低氧缺血新生鼠 (n =8)不同脑组织内皮素 (ET)的变化。 (2 )低氧缺血后不同时间 (即刻、6 0及 12 0min ,n分别为 9,8,8)新生鼠脑内ET的变化及与脑水肿的关系。 (3)低氧缺血新生鼠 (n =11)脑内ET、丙二醛及兴奋性氨基酸的变化及应用ET抗血清 (n =11)后的可能的变化。结果 (1)正常新生鼠不同脑区ET呈不均匀分布 ,垂体含量最高。低氧缺血组 2h后垂体ET含量为 (41.7± 10 .8)pg/mg],大脑皮层次之 [(33± 7)pg/mg]。(2 )低氧缺血新生鼠大脑皮层ET含量明显增高 ,并与脑水肿呈显著正相关 (r=0 .95 4,P <0 .0 1)。 (3)与对照组比较 ,低氧缺血时脑组织丙二醛、谷氨酸、天门冬氨酸及ET明显升高 [分别为 (2 0 8± 19)nmol/g、(10 .7± 1.7)nmol/mg、(3.9± 0 .8)nmol/mg、(6 9.7± 10 .2 )pg/mg],而ET抗血清能有效抑制脑丙二醛、天门冬氨酸及ET的产生 [分别为 (133± 2 2 )nmol/g、(2 .9± 0 .3)nmol/mg、(2 0 .2± 3.6 )pg/mg]。结论 ET参与低氧缺血脑损伤的发病过程 。
Objective To explore the possible role of anti-endothelin (ET) serum in neonatal rats with hypoxic -ischemic brain damage. Methods The changes of brain tissue ET, malonicaldehyde (MDA) and excitatory amino acids and possible actions of anti-ET serum in 7 neonatal rats with hypoxic-ischemia and 8 control rats were observed by RIA, TBA and amino acid autoanalysis. Results (1) There were different distributions of ET contents in different brain regions of normal neonatal rats. The highest level was in pituitary. The contents of ET after hyponic-ischemia 2 hours were in the pituitary (41 7±10 8) pg/mg, and (33±7) pg/mg in hypothalamus. (2) Cortex ET contents in neonatal rats with hypoxic-ischemia were significantly higher than those in control rats, and correlated significantly with cerebral edema ( r =0.954, P <0.01). (3) Compared with those in controls, the brain tissue MDA, Glu, Asp and ET levels in hypoxic-ischemic group increased obviously (208±19) nmol/g, (10.7±1.7) nmol/mg, (3.9±0.8) nmol/mg and (69.7±10.2) pg/mg, respectively. Anti-ET serum could effectively inhibit the productions of MDA, Asp and ET (133±22) nmol/g, (2.9±0.3) nmol/mg and (20.2±3.6) pg/mg, respectively. Conclusion ET was involved in the development of hypoxic-ischemia in neonatal rats. Anti-ET serum could protect the neonatal rats from hypoxic-ischemic brain damages.
