摘要
目的:选用体外培养人多形性胶质母细胞BT325作为对象,研究抗癌药物VM-26的治疗作用机理。方法:采用荧光显微镜、电子显微镜、激光扫描共聚焦显微镜、流式细胞分光光度计和DNA凝胶电泳等方法和技术观察细胞凋亡。结果:实验显示VM-26作用于体外培养人多形性胶质母细胞BT325后可诱导其发生凋亡。结论:VM-26抑制体外培养BT325细胞的生长,是通过阻滞细胞周期的进程,启动细胞自身的调控程序,诱导细胞凋亡,从而达到抗肿瘤的作用。
Objective: The purpose of this experiment is to investigate the mechanism of therapeutic effect of anticancer drug teniposide on cultured human glioblastoma multiforme cells BT325. Method: Using fluorescence microscopy, electron microscopy, laser scanning confocal microscopy, flow cytometry and DNA gel electrophoresis to demonstrate the presence of apoptosis. Result: The results of this experiment indicate that the therapeutic effect of VM-26 on BT325 cell was induced by apoptosis. Conclusion: The mechanism of apoptosis is involved in the inhibition of BT325 cell growth caused by VM-26 treatment.
