红景天甙对肝癌细胞增殖及斑马鱼血管生成作用的研究 被引量:10

摘要 目的:探讨红景天甙对鼠肝癌细胞(W256,H22,Hepa 1-6)增殖及对斑马鱼血管生成以及对人脐静脉内皮细胞(HUVEC)生长及迁移的影响的作用。方法:研究不同浓度红景天甙作用下肝癌细胞(W256,H22,Hepa 1-6)的增殖情况。采用24hpf(hours post fertilization,hpf,受精后小时)健康TG(VEGRR2/GFP)系血管荧光转基因斑马鱼作为实验动物模型,加入不同浓度红景天甙与胚胎共同孵育后在荧光显微镜下观察背部节间血管(intersegmental vessels,ISV)生成数目。研究红景天甙对人脐静脉内皮细胞(HUVEC)生长及迁移的影响。结果:红景天甙抑制肝癌细胞(W256,H22,Hepa 1-6)的增殖,并能有效抑制斑马鱼血管生成及抑制HUVEC的生长及迁移,且不显示明显细胞毒性。结论:红景天甙可明显抑制肝癌细胞增殖,并能抑制血管生成,具有研究开发价值。 Objective:To investigate the effect of salidroside on proliferation of mouse hepatoma cell(W256,H22,Hepa 1-6) and angiogenesis of zebrafish.Methods:To explore the proliferation of hepatoma cell(W256,H22,Hepa 1-6).Healthy transgenic zebrafish TG(VEGRR2/GFP) at 24hpf was used as animal model.Growth of intersegmental vessels(ISV) of larva trunk was scored under fluorescence microscope after different concentrations of salidroside were added into medium of zebrafish and fertilized with zebrafish embryos for 36h.Results:The salidroside could inhibit the proliferation of hepatoma cells(W256,H22,Hepa 1-6) and the angiogenesis of zebrafish,it could also inhibit the growth and migration of HUVEC.Conclusion:The salidroside could inhibit the proliferation of hepatoma cells significantly,and it could to exert their anti-tumor efficacy by inhibiting angiogenesis.
