银杏叶提取物对重症急性胰腺炎大鼠一氧化氮及诱导型一氧化氮合成酶的影响 被引量:1

摘要 目的探讨银杏叶提取物(GBE)对重症急性胰腺炎(SAP)大鼠模型一氧化氮(NO)及诱导型一氧化氮合成酶(iNOS)表达的影响。方法 72只SD大鼠随机分为3组:假手术组(SO)、SAP组、GBE干预组。采用5%牛磺胆酸钠胰胆管注射方法建立SAP动物模型。GBE治疗组于建模后5 min将GBE溶液通过股静脉给药。SO组和SAP组注入同样量的生理盐水。检测3组6、12、24 h血清淀粉酶、胰腺组织病理评分、血清及胰腺组织NO水平、实时荧光定量PCR检测胰腺组织iNOS mRNA表达情况。结果 GBE组胰腺组织病理评分6、12 h均显著低于SAP组,但在24 h胰腺组织病理评分与SAP组无显著差异;GBE组在6 h时血清及胰腺组织NO水平、12 h时胰腺NO水平较SAP组低,但随着时间延长,两组无显著差异;6 h时GBE组iNOS mRNA表达较SAP组低(P<0.05);12、24 h时SAP组与GBE组iNOS mRNA表达无统计学意义。结论 GBE在SAP早期可能通过抑制iNOS mRNA表达而降低NO水平,对胰腺组织起保护作用;但随着病程进展,这种作用不明显。 Objective To research the effects of ginkgo biloba extract (GBE) on nitric oxide level and expression of inducible nitric oxide synthase in severe acute pancreatitis in rats. Methods A total of 72 SD rats were divided randomly into three groups: sham operation group (SO), severe acute pancreatitis group (SAP) and GBE group. SAP model was established by intraductal retrograde injection of 5% sodium taurocholate. In GBE group, GBE solution was injected 5 min after the oper- ations by femoral vein,while the SO group and the SAP group received an infusion of saline of the same dosage. The rats were sacrificed at 6 h, 12 h and 24 h respectively,and their blood samples were collected for analyses. The variation of serum amylase levels and the pathohistological changes of the pancreas were detected by colorimetric method. The level of NO in serum and pancreas were tested by nitrale reduetase. Experssion of iNOS mRNA in the pancreas were determined by real-time fluorescent quantitative PCR. Results The pancreas pathological score of GBE group were lower than SAP group at 6 h, 12 h, but there was no difference at 24 h subgroup. The NO level of serum and pancreas at 6 h, the NO level of pancreas at 12 h were lower in the GBE group. But as time goes by, there were no differences. The expression of iNOS mRNA was lower in the GBE group at 6 h(P 〈 0.05), but there were no differences between the GBE group and SAP group at 12 h, 24 h. Conclusion At an early stage of SAP, GBE may restrain the iNOS expression,and then reduce the NO level, play a key role of pancreas tissue protection. However, as time goes by, the protection is not significant.
