摘要
目的观察醒脑静微乳经鼻给药后对大鼠行为及鼻黏膜形态学的影响,探索开发新型复方制剂。方法健康SD大鼠125只,随机分为空白微乳组、空白溶剂组、醒脑静微乳组(载药量为84.4g/L)、醒脑静滴鼻液组(载药量为84.4 g/L)以及生理盐水组,每组25只,各组每日给药3次,每次每鼻孔定量注入50μL相应液体,连续给药7 d,于给药1、4、7 d和停药1、2周后,取鼻中隔黏膜,制备石蜡切片,HE染色,光镜下观察,并对大鼠进行行为学观察。结果各组鼻腔给药后,大鼠有不同程度的行为反应。切片观察可知空白微乳组、空白溶剂组与醒脑静微乳组给药7 d后对大鼠鼻黏膜有一定的刺激性,且刺激性相当(P>0.05),但损伤于停药1周后即恢复正常;醒脑静滴鼻液组给药7 d后对大鼠鼻黏膜刺激性显著高于微乳组(P<0.05)。结论醒脑静中成分对大鼠鼻黏膜有较大的刺激性,但将其制成微乳后可减少药物对鼻黏膜的刺激性且损伤于1周后恢复正常。
Objective To observe the influence of Xingnaojing microemulsion after nasal administration on rats' behaviors and mo Methods Health SD rats rphology of nasal mucusa, and develop new kind of compound preparation. (n = 125 ) were randomly divided into blank microemulsion group (blank group 1 ), blank solvent group (blank group 2), Xingnaojing microemulsion group (Xingnaojing group 1, 84.4 g/L), Xngnaojing naristillae group (Xingnaojing group 2, 84. 4 g/L) and normal saline solution group (saline group, each n =25). Each group was given corresponding medicinal (50 txL) to every nostril each time and three times a day for 7 days. The samples of nasal septum mucosa were collected respectively after 1, 4 and 7 days and 1 and 2 weeks after stoping medicinal administration for preparing paraffin sections, which were given HE staining and observed under light microscope. The ethological changes were observed in rats. Results After nasal administration of medicinals in all groups, the rats had behavioral reactions in varying degrees. The results of section observation showed that blank group 1, blank group 2 and Xingnaojing group 1 had certain irritating effect on nasal mucosa 7 days after medicinal administration and the irritation was equivalent (P 〉0.05 ), but the injury of nasal mucosa was recovered one week after stopping medicinal administration. The irritation to nasal mucosa was significantly higher in Xingnaojing group 2 than that in Xingnaojing group 1 ( P 〈 0.05 ) after 7 days. Conclusion The medicinals in Xingnaojing have irritation to nasal mucosa in rats, while when Xingnaojing is prepared as microemulsion, the irritation is reduced and the injury of nasal mucosa is recovered after one week.
