摘要
以照山白杜鹃(Rhododendron micranthum)试管苗的茎段为外植体,研究不同激素组合对其外植体诱导、丛生芽增殖及试管苗生根的影响。结果表明:最适丛生芽诱导培养基为Read+ZT 4.0 mg/L+NAA 0.05 mg/L+蔗糖(3%),诱导率达92.41%;最适丛生芽增殖培养基为Read+ZT 3.0 mg/L+NAA 0.1 mg/L+蔗糖(3%),增殖系数达7.56;生根培养基为Read+IAA 0.5 mg/L+蔗糖(2%),生根率达92.5%;NAA不适宜照山白杜鹃生根诱导;将试管苗移栽到松毛土:泥炭土:河沙=1:2:1的基质中,成活率达89%。
The stems of plantlet were used as explants to study the effect of different hormone combinations on explant induction, propagation of clustering shoots and the rooting of Rhododendron micranthum. The results indicated that the best medium for shoot induction was Read+ ZT 4.0 mg/L+NAA 0.05 rag/L+ sugar (3%), with the inductivity rate of 92.41%. The best medium for multiplication was Read+ZT 3.0 rag/L+ NAA 0.1 mg/L + sugar (3%), with multiplication coefficient of 7.56. Read+IAA 0.5 mg/L +sugar (2%) was the best medium for rooting, with rooting rate up to 92.5%. At same time, NAA was proved not to be useful for root induction in R. micranthum. The survival rate reached 89% when the planflets were transplanted in substrate ofturfy soil: humus soil: riversand = 1: 2: 1.
