摘要
用三种杜鹃花的茎尖组培快速繁殖,主要对增殖培养中激素种类与浓度进行了较深入的研究。根据本试验的增殖系数和培养时间推算,一个茎尖一年内可获得试管苗15000~20000株。
Rapid propagation of shoot tips of three species of Rhododendron spp.was studied by adjusting the kinds and concentrations of hormones applicd to R. pulchrum,R.simsii and R.mucronulatum,2ip or Zeatin(ZT)was used for R. simsii and R.mucronulatum,but ZT for R.pulchrum.An average of 5 to 8 fold multiplication rate of shoots and an 8 to 12 fold multiplication rate of itiate stem sections of three rhododendron species were produced after 1.5 months of culture.The harvested shoots were rooted in R+IBA at 5mg/1+0.1% activated charcoal;the rooting rate was about 60% after 1.5 months of culture. Three rooting methods were used.Plantlets were first rooted in vitro,then cutting were protreated with IBA at 10mg/l for 10 days and planted,and fnially cuttings were dipped in IBA at 100ppm for 10 seconds and planted.The medium for cuttings or plantlets was 1 peat:1 vermiculite.The survival rates for the three methods were all abotut 70%.However,the third method is most convenient in terms of handling.According to a preliminary estimate, 15000-20000 rooted plantlets could be obtained from cone shoot tip of three species of rhododendron after one year.
