摘要 目的重组质粒转染HeLa细胞以探讨Grim-19表达水平的变化对HeLa细胞中端粒酶活性的影响。方法将重组质粒p3×flag-myc-cmv-24-Grim-19(简写Grim-19)通过脂质体转染至HeLa细胞,48 h后通过Western blot检测Grim-19、htert、P16、P21的表达情况,同时检测HeLa细胞衰老和端粒酶活性。结果转染后可见flag的表达及htert的表达降低及P16、P21的表达升高,细胞衰老增加,增殖减弱及端粒酶活性的降低。结论在子宫颈癌细胞株HeLa中Grim-19具有调控端粒酶活性和细胞衰老的作用。 Objective To study the effect of the changing expression of Grim-19 on the telomerase activity in HeLa cell which was transfected from recombinant plasmid. Methods After the plasmid p3 × flag-myc-cmv-24-Grim-19 (Grim-19) was transfeeted into the HeLa cells, Western blot were used to test the expression levels of Grim-19, htert, P16 and P21 after 48 h. The senescence and telomerase activity of HeLa cell were also detected. Results The low expression of flag and htert and overexpression of P16 and P21 can be obvious after the transfection, and the increasing cellular senescence and weakening proliferation and reducing telomerase activity were also found to be- true. Conclusion Grim-19 can regulate the telomerase activity and cell senescense in the cervical cancer cell line.
