摘要
为了石榴遗传转化研究的开展,以"突尼斯软籽"石榴为研究材料,叶片作为外植体,研究了不同灭菌时间和不同生长调节物质对离体叶片愈伤组织再生的影响。结果表明:最适宜的暗培养时间为14 d,最佳愈伤组织分化培养基为MS+BA 1.0 mg.L-1+NAA(0.3~0.5)mg.L-1,最佳不定芽分化培养基为MS+NAA 0.3 mg L-1+TDZ 1.5 mg L-1+KT(1.5~2.0)mg L-1,最佳新梢增高的培养基为MS+NAA 1.0 mg L-1+BA 0.2 mg L-1+KT 1.0 mg L-1,适宜生根的培养基为MS+NAA0.5 mg L-1,生根率达100%。
In order to develop genetic transformation test of pomegranate, using leaves in Tunisia soft-seed pomegranate as explants, the effects of different sterilization time and different growth regulators on leaf regeneration in vitro were studied. The results showed that the optimum dark culture time was 14 days. The optimum medium for callus differentiation was MS supplemented with BA 1.0 mg-L1 and NAA (0.3-0.5) mg'L1. The optimum medium for adventitious buds differentiation was MS supplemented with NAA 0.3 mg·L-1, TDZ 1.5 mgoL1 and KT (1.5-2.0) mgoL1. The optimum medium for shoot growth was MS supplemented with NAA 1.0 mg·L-1, BA 0.2 mg·L-1 and KT 1.0 mg·L-1. The best medium for rooting was MS supplemented with NAA 0.5 mg·L-1, and rooting rate reached 100%.
