摘要
为了开展石榴遗传转化研究,以‘突尼斯软籽’石榴为试材,叶片为外植体,采用响应面回归分析方法研究了不同生长调节物质对离体叶片愈伤组织再生的影响。结果表明,‘突尼斯软籽’石榴叶片愈伤组织诱导的最佳培养基为MS+6-BA 1.5 mg/L+2,4-D 0.5 mg/L+NAA 0.3 mg/L,诱导率可达83.9%,且愈伤组织生长状况良好。本研究建立了‘突尼斯软籽’石榴高效的离体快繁体系,为石榴繁育技术的改善提供了理论依据。
In order to develop genetic transformation test of pomegranate,in this paper,using leaves in‘Tunisia soft-seed’pomegranate as explants,the different growth regulators on callus regeneration of leaves by response surface method was studied.The results showed that the optimal medium was MS+1.0 mg/L 6-BA+1.0 mg/L 2,4-D+0.2 mg/L NAA,the induction rate was 83.9%,and most of the callus grew well.At the same time,a high efficient rapid propagation system of‘Tunisia soft-seed’pomegranate was established.And it also provided a theoretical basis for improving pomegranate breeding technology.
