摘要
[目的]优化派间杂种黑白杨的再生和遗传转化体系。[方法]以黑白杨3种不同基因型组培苗叶片及茎段为材料,采用正交试验研究了不同激素配比、基因型及外植体状态对其生根、侧芽生长及愈伤组织再生的影响。[结果]1/2MS+IBA0.4mg/L+NAA0.1mg/L对黑白杨的4号基因型是最优的生根培养基;MS+6-BA0.8mg/L+NAA0.2mg/L是诱导黑白杨茎段侧芽生长的最优培养基;MS+6-BA1.0mg/L+NAA0.1mg/L是诱导黑白杨叶片再生不定芽生成的最适培养基。[结论]建立了完整的黑白杨再生体系。
[Objective] The regeneration from the hybrid between the cotton-wood and aspen and its genetic transformation system were optimized.[Method]The leaf and stem of 3 different genotypes of plantlets from the hybrid between cotton-wood and aspen being taken as the experimental material,the effect of the hormone,genotype and explants on the rooting,the lateral bud growth and the regeneration from callus was studied with the design of orthogonal experiment.[Results] The best rooting medium for the genotype No.4 from the hybrid between cotton-wood and aspen was 1/2 MS + IBA 0.4 mg/L + NAA 0.1 mg/L;the best suitable medium for the bud lateral bud growth from the stem of the hybrid between cotton-wood and aspen was MS + 6-BA 0.8 mg/L + NAA 0.2 mg/L;the best medium for the adventitious bud growth from the leaf of the hybrid between cotton-wood and aspen was MS + 6-BA 1.0 mg/L + NAA 0.1 mg/L.[Conclusion] The regeneration system from the hybrid between cotton-wood and aspen was established.
