人参内生可培养细菌16S rDNA-RFLP分析 被引量:10

16S rDNA-RFLP Analysis of Culturable Endophytic Bacteria from Panax ginseng

摘要 以我国吉林省5个不同地区来源的健康人参植株为实验材料,分别从根茎叶不同器官分离获得了152株内生细菌,利用HhaⅠ、HinfⅠ和HaeⅢ3种限制性内切酶对所有供试菌株的16 S rDNA片段进行了RFLP分析。聚类分析结果表明,供试菌株在相似性为83%水平上可分为18个类群,不同地区内生细菌的遗传多样性存在差异,其中抚松县和吉林农业大学的菌株多样性较高;不同器官蕴含的菌株数目也存在差异,茎部和根部蕴含菌株较为丰富。研究结果期望为了解人参内生细菌的种群结构,明确菌株种类奠定基础。 Healthy Panax ginseng plants from five different areas in Jinlin Province of China were selected. A total of 152 strains of endophytic bacteria were isolated from roots, stems and leaves of ginseng respectively. The 16S rDNA-RFLP was analyzed with three types of restriction enzymes of Hha I , Hinf I and Hae III. Results showed that all strains were clustered into 18 groups at the genetic similarity level of 83%. The genetic diversities of endophytic bacteria isolated from the five regions and three organs were all different, in which the diversities of strains from Fusong County and Jilin Agricultural University were higher, and the strains from stems and roots were abundant.
