摘要
目的 :探索用人羊膜作为胚胎着床模型支持材料的可行性。方法 :将妊娠第四天的小鼠囊胚培养在人羊膜的基膜面 ,并作病理观察。结果 :小鼠囊胚能在羊膜上正常脱带、贴附和扩展 ,其脱带率和贴附率与在培养板中的结果没有差异 ,但扩展率显著低于在培养板中的值 ;滋养层细胞对羊膜的基膜有侵入 ,二者在结构上有交错。结论 :人羊膜可用作胚胎体外着床模型的建立 。
Objective: To develop a new embryo implantation model with human amnion. Methods:Culture the mouse blastocysts on day 4 of pregnancy on human amniotic basement membrane and observe microscopically. Results :Mice blastcysts developed normally experiencing shedding zona pellucida,attachment and outgrowth.There was no difference between the percentage of blastocysts shedding zona pellucida and attachment cultured on the membrane and in the 96-well plates while the percentage of outgrowing blastocycts on the membrane was significantly lower than that in the plates.However,light microscopic examination indicated that trophoblasts invaded the basement membrane while it was impossible to do so on the plastic or glass surfaces. Conclusion: Human amnion could be used as substratum of in vitro model for peri-implantation development of mouse blastocysts.
