摘要
目的:探讨三氧化二砷(As_2O_3)诱导大鼠血管平滑肌细胞(VSMCs)调亡的作用。方法:建立大鼠VSMCs增生模型。用四甲基偶氮唑蓝还原反应(MTT法)、透射电镜、琼脂糖凝胶电泳、流式细胞术等方法观察 As_2O_3诱导 VSMCs凋亡的作用。结果:经As_2O_3作用后VSMCs出现典型的凋亡变化,透射电镜下可见细胞核仁消失,染色质浓缩聚集于核膜下。DNA琼脂糖凝胶电泳出现凋亡特征性梯形电泳条带。流式细胞术检测出现典型的凋亡峰,并随AS_2O_3浓度的增加和作用时间的延长而增高。对照组均不见上述变化。结论: As_2O_3可有效诱导 VSMCs凋亡。
Objective: To study the apoptotic effect of arsenic trioxide (As_2O_3, ) on vascular smooth muscle cells (VSMCs) in rats. Methods: VSMCs proliferation model of cultured rat thoracic aorta was established. And NBT reduction test, transmission electron microscopy, agarose gel electrophoresis and flow cytometry were applied to observe the apoptotic changes of VSMCs induced by As_2O_3. Results: Typical apoptotic cellular changes were found after As_2O_3 treatment, transmission electron microscopy showed nucleole disappeared, chromatin condensated under nucleonic membrane. Agarose gel electrophoresis showed 'ladder' strand of DNA, a special phenomenon of cellular apoptosis. And an AP peak of degraded DNA could be seen by flow cytometry, which heightened in a concentration: and time-dependent manner. The above- mentioned changes were not found in the control group. Conclusion: As_2O_3 can induce apoptosis of rat proliferated VSMCs effectively.
