摘要
目的探讨三氧化二砷(As2O3)治疗白血病的作用机制。方法在K562细胞中表达带3蛋白膜段结构域,经WesternBlot检定其表达;通过离子转运测定验证其生物学活性;应用As2O3诱导细胞凋亡,通过电镜观察、线粒体膜电位测定和凋亡片断DNA的提取,观察As2O3对表达带3蛋白膜段结构域的K562细胞凋亡的影响。结果表达带3蛋白膜段结构域的K562细胞的凋亡特征比未表达带3蛋白的K562细胞明显,说明表达带3蛋白膜段结构域的K562细胞对As2O3更敏感。结论带3蛋白参与了凋亡的过程,为探讨As2O3治疗白血病的机制提供一条新的线索。
[Objective] To investigate the mechanism of Arsenic Trioxide treatment to Leukemia. [Methods]We expressed band3 membrane domain in K562 cells which was identified by Western blot. The effect of As203 on K562 cells which expressed band3 membrane domain was observed with TEM, mitochondrial membrane potential (MMP)assay and DNA electrophoresis. [Results] As2O3 induced more significant changes consistent with apoptosis in K562 cells expressed band3 membrane domain. [Conclusion] The K562 cells expressed band3 membrane domain were more susceptible to As2O3, which provided a new clue to study the mechanism of arsenic trioxide treatment to leukemia.
