反相高效液相色谱内标法测定儿童血清中视黄醇的含量 被引量:3

摘要 目的建立反相高效液相色谱内标法测定儿童血清中维生素A的方法,以评价湖北省十堰市3~9岁儿童维生素A营养状况。方法取血清加入乙醇及正己烷萃取,涡旋混合,离心后,移取己烷层氮气吹干,残渣加入无水乙醇溶解进样。色谱条件:流动相:甲醇:水(90:10 v/v),柱温:25℃,流速:1 ml/min,检测波长325 nm,内标法定量。结果维生素A在浓度(0.218 2~6.982)mol/L范围内呈线性,平均回收率为100.1%,全部操作可在20 min内完成。测得这些儿童血清视黄醇均数为(1.48±0.52)mol/L,维生素A缺乏率为3.0%,边缘缺乏率为13.6%。结论该方法快速、灵敏、准确,适合大样本调查时使用。十堰市儿童维生素A营养状况与2002年的全国平均水平相比较好,仍应注重对儿童维生素A边缘性缺乏的预防。 Objective A method for determination of sentm retinol concentration of children by HPLC was developed to evaluate the vatamin A nutritional status of children aged 3 - 9 years old in Shiyan city. Methods Vitamin A was extracted by N-hexane, after vortexed and centrifuged, the hexane layer was moved and dried by nitrogen evaporation, dissolved by absolute ethanol, then analyzed by HPLC. Retinol was detected at 325nm with UV detector by using methanol/water (90:10 v/v) as mobile phase, the column temperature was 30~C, and the flow rate was lml/min. Quantity was determined by internal standard method. Re.suits The linear range was 0.218 2 -6.982 mol/L. The recoveries were 100. 1%. The determination was completed within 20 minutes. The mean ( ± SD) serum retinol concentration was 1.48 ±0. 52 mol/L,the prevalence of vitamin A deficiency (VAD) was 3.0%, the prevalence of marginal VAD was 13.6%. Conclusions The method was rapid, sensitive and accurate. It can be used when large sample survey was performed. Although the nutritional status was better than the average level of the national survey in 2002, the prevention of marginal YAD of children in Shiyan city should be further emphasized and improved.
