摘要
目的探讨食管鳞状细胞癌(ESCC)中WIF-1基因启动子甲基化状态及mRNA表达水平及其与病理指标之间的关系。方法应用甲基化特异性聚合酶链反应(MSP)技术和逆转录-聚合酶链反应(RT-PCR)技术检测62例ESCC组织及30例癌旁组织中WIF-1基因启动子区甲基化状态和mRNA的表达水平,应用SPSS13.0探讨其与临床病理资料的关系。结果 WIF-1基因在癌组织中的甲基化率59.7%(37/62)明显高于癌旁组织13.3%(4/30),差异有显著性(P<0.05);其甲基化状态与年龄、分化程度及淋巴结转移情况相关(P<0.05);而与其他临床资料(性别、部位、浸润深度、临床分期)无明显相关性(P>0.05)。WIF-1基因mRNA在ESCC组的表达量(0.565±0.112)显著低于癌旁正常组(0.666±0.107),差异有显著性(P<0.05)。结论 WIF-1基因启动子甲基化在食管鳞癌的进展过程中起着重要的作用;WIF-1基因mRNA表达异常可能与ESCC的发生有关,此基因甲基化与mRNA表达两者之间的关系有待进一步研究。
【Objective】 To investigate the hypermethylation status and mRNA expression of WIF-1 gene in esophageal squamous cell carcinoma(ESCC).【Methods】 Methylation status of WIF-1 gene promoter were detected by methylation specific PCR in 62 esophageal squamous cell carcinoma and 30 adjacent non-cancerous tissues.Expression of WIF-1 mRNA was also determined by reverse transcriptase PCR(RT-PCR) in these tissues.The relationship between methylation of the gene and clinical data were analyzed.【Results】 The methylation rate of the ESCC and corresponding non-tumor tissues were 59.7% and 13.3% respectively.Methylation of the WIF-1 gene in ESCC increased significantly compared with adjacent non-tumor tissues(P〈0.05);there was significant difference in age,differentiation and the metastasis of lymph node in the tumor tissues;there was no significant difference in other clinical data(sex,age,infiltration,clinical stage) of the tumor;expression of WIF-1 mRNA in adjacent non-cancerous tissues(0.666±0.107) was significantly higher than in esophageal squamous cell carcinoma(0.565±0.112).【Conclusion】 The promote methylation of WIF-1 gene took an important role in the process of the ESCC,abnormal expression of the WIF-1 mRNA may be related with oncogenesis of ESCC,and the relationship between them will further study.
