肝细胞癌中WIF-1基因启动子甲基化状态及mRNA表达的研究 被引量:3

PROMOTER METHYLATION AND MRNA EXPRESSION OF WIF-1 GENE IN HEPATOCELLULAR CARCINOMA

摘要 目的探讨细胞癌中WIF-1基因启动子甲基化状态及其对mRNA表达的影响。方法应用甲基化特异性聚合酶链反应和实时定量逆转录-聚合酶链反应技术,检测33例肝细胞癌及相应的癌旁组织和20例正常对照肝组织中WIF-1基因启动子甲基化状态和mRNA表达水平,分析上述组织WIF-1基因甲基化及mRN表达与临床资料的关系并分析甲基化对mRNA表达的影响。结果WIF-1基因在癌组织中甲基化率明显高于癌旁组织和正常组织(χ2=4.89,P<0.05),而癌旁组织和正常组织中WIF-1基因甲基化率相比无明显差异;癌组织中WIF-1基因甲基化与乙肝表面抗原、肝硬化有关;WIF-1基因mRNA表达量在三种组织中比较无明显差异。结论WIF-1基因启动子甲基化与HCC的发生相关;WIF-1基因启动子甲基化与其mRNA表达的关系有待进一步研究。 Objective To investigate the promoter methylation status and mRNA expression of WIF-1 gene in hepatocellular carcinoma. Methods Methylation status of WIF-1 gene promoter were detected by methylation specific PCR in 33 hepatocellular carcinomas and adjacent non-cancerous tissues,20 normal liver tissue as control. Expression of WIF-1 mRNA was also determined by Real Time Quantitative reverse transcriptase PCR in the three tissues. The relationship between methylation and clinical data, mRNA expression and clinical data, methylation and mRNA expression of the genes were analyzed. Results Methylation of WIF-1 genes in hepatocellular carcinoma increased significantly compared with adjacent non-cancerous tissues and normal control tissues (Х^2 = 4. 89, P 〈 0. 05) and there was no significant difference in methylation between adjacent non-cancerous tissues and normal control tissues ; significant differences of the WIF-1 promoter methylation were observed in HBsAg and cirrhosis ; Average expression of WIF-1 mRNA was no significant difference among three tissues. Conclusion The aberrant promote methylation of WIF-1 genes are both related to hepatocarcinogenesis and progress. The relationships between methylation and mRNA expression of the two genes will further study.
