摘要
目的:观察乌梅丸拆方对溃疡性结肠炎(UC)大鼠的治疗作用,探讨不同治法的作用机制及该方寒热并用在治疗溃疡性结肠炎中的配伍意义。方法:以UC大鼠为研究对象,ELISA测结肠组织细胞因子(TNF-α等)及炎性因子(PGE2等)含量,比色法检测结肠组织和血清SOD活性,测算脾指数、结肠指数等。结果:与模型组比较,乌梅丸组、寒热并用组大鼠死亡率、体温、脾指数均降低,体质量升高,结肠组织TNF-α、IL-1、PGE2含量减少(P<0.05),IL-10含量增多(P<0.05);温热组结肠组织PGE2、TNF-α、IL-1含量减少(P<0.05),IL-10含量增多(P<0.05);寒凉组大鼠体温、脾指数降低,结肠组织TNF-α、IL-1含量减少(P<0.05);补益组大鼠死亡率、脾指数、体温降低,体质量升高,结肠组织TNF-α、IL-1含量减少(P<0.05)。结论:寒热配伍是乌梅丸方中治疗溃疡性结肠炎的主要配伍形式,其主要机制为调节细胞因子间平衡,减少炎性介质等;乌梅丸各拆方通过不同的作用机制对溃疡性结肠炎起到治疗作用。
Objective： To observe the treatment effects of Wumei Pill Subdivisions on rats of ulcerative colitis（UC） and discuss the mechanism of different therapeutic methods,and the compatibility significance of application of both cold and heat in Wumei Pill on treat ulcerative colitis（UC）.Methods： Using UC rats as the research object,the content of cell factor such as TNF-α、IL-1、PGE2 were determined in rats colon tissue with ELISA method,and the SOD activity in rats colonic tissue and serum with colorimetric measurement,calculate the index of spleen and colon were determined as well.Results： Compared with the model group,the mortality,body temperature,and spleen index of rats all decreased while the weight of rats increased,and the content of TNF-α、IL-1、PGE2 in rats colonic tissue all decreasd significantly（P0.05） while the IL-10 increased（P0.05） in the Wumei Pill group and cold and heat application group;the content of TNF-α、IL-1、PGE2 in rats colonic tissue all decreased（P0.05） while the IL-10 increased（P0.05） in the thermal group;the body temperature,spleen index and the content of TNF-α、IL-1 all decreased（P0.05） in the cold group;the mortality,body temperature,spleen index and the content of TNF-α、IL-1、PGE2 in rats colonic tissue all decreased（P0.05） while the weight of rats increased in tonic group.Conclusion： The compatibility of cold and heat is the main compatibility form of Wumei Pill in the treatment of ulcerative colitis.One of the main mechanisms is the regulation of cytokine balance,and reduces the inflammatory mediators.Wumei Pill Subdivisions have an effect on treating ulcerative colitis by different mechanisms.
