摘要
目的:建立五味清浊散中羟基红花黄色素A的高效液相色谱测定方法。方法:色谱柱为Prevail Select C_(18)(150 mm×4.6 mm,5μm),柱温35℃;流动相为乙腈-0.5%磷酸溶液(11:89),流速1 ml·min^(-1);检测波长403 nm。结果:羟基红花黄色素A在16.65~166.50μg·ml^(-1)范围内呈良好的线性关系,回归方程为Y=-3.24×10^(-2)X+2.12,r=0.999 9。平均回收率为99.1%,RSD为1.9%。结论:本方法操作简便,测定结果准确可靠,可用于五味清浊散中羟基红花黄色素A的含量测定。
Objective: To establish an HPLC quantitative method for the determination of hydroxysafflor yellow A in Wuwei Qingzhuo powders. Method: High Performance Liquid Chromatography was used for the determination of hydroxysafflor yellow A. The chromatography column was Prevail Select C18 (150 mm × 4.6 mm,5 μm)and the column temperature was 35℃. The mobile phase was acetonitrile-0.5 % phosphoric acid ( 11: 89). The flow rate was 1 ml ·min^-1 and the detection wavelength was at 403 nm. Result: The retention time of hydroxysafflor yellow A was about 14 min with resolution above 1.5. The regress equation for hydroxysafflor yellow A was as the following : Y = 3.24 × 10^ -2X + 2.12 ( r = 0.999 9), and the linear range was 16.65-166.50 μg · ml ^- 1. The average recovery of hydroxysafflor yellow A was 99.1% ( RSD = 1.9% ). Conclusion : This method is simple, time-saving and accurate, It can be used for the routine analysis of hydroxysafflor yellow A in Wuwei Qingzhuo powders.
