摘要
目的 探讨"合欢皮298"对小鼠肝癌血管生成的作用.方法 建立皮下肝癌移植瘤模型的20只小鼠随机分成2组,治疗组建模后24 h使用"合欢皮298"5 mg/(kg·d)灌胃治疗,连续7天,对照组给予等量生理盐水.在服药后7天和14天分别对小鼠进行常规超声及实时灰阶超声造影(contrast-enhanced ultrasonography,CEUS)检查,使用Sonoliver软件获得造影时间强度曲线(time intensity curve,TIC),比较TIC参数的差异.于服药后14天超声造影检查完成后处死小鼠,完整剥离肿瘤,免疫组化检测肿瘤CD31表达情况,进行微血管密度(micro vessel density,MVD)分析.结果 治疗组肿瘤体积小于对照组(P<0.05);7天、14天治疗组小鼠CEUS定量参数最大峰值强度(maximum intensity,IMAX)小于对照组(P=0.000);7天时2组间上升时间(rise time,RT)、达峰时间(time to initial peak,TTP)、平均渡越时间(mean transit time,mTT)差异无统计学意义,14天时治疗组RT、TTP、mTT大于对照组(P〈0.05);治疗组和对照组MVD差异有统计学意义(P=0.009).结论 "合欢皮298"具有抑制肿瘤血管生成的作用,有望成为有效的肿瘤血管生成抑制剂.
Objective To investigate the effect of Albizia julibrissin 298 on angiogenesis of hepatoeellular carcinoma in mice. Methods 20 mice were subcutaneously implanted with H22 cells. 10 mice were treated with Albizia Julibrissin 298 (5 mg. kg-1, d-1 ) po. over 7 days. 10 control mice were treated with an equivalent volume of normal saline. Volume measurement and CEUS were performed on day 7 and day 14, respectively. Regions of interest within tumour and nomal tissue were analyzed off - line with the software of Sonoliver to obtain time - intensity curve (TIC), and determine perfusion parameters such as the maximum intensity( IMAX), rise time (RT), time to initial peak (TI'P), mean transit time (mTT). On day 14, mice were killed and tumour tissue was removed immediately after imaging. Microvascular density (MVD) was measured after anti - CD31 staining. Results The tumour volume of treatment group was significantly different than that of control group ( P 〈 0.05 ) ; There were significant difference in IMAX compared with treatment group and control group (P 〈 0.05 ) on day 7 and day 14; No statistically significant difference was observed in RT ,TTP and mTT between treated and control group on day 7, while there was significant difference in RT ,TTP and mTT on day 14 ( P 〈 0.05 ). Treated tumours with a significantly lower MVD as compared with control tumours (P 〈 0.01 ). Conclusion Albizia julibrissin 298 exerts antiangiogenesis effect on hepatocellular carcinoma in mice, which is expected to become a effctive angiogenesis inhibitor.
