摘要
目的建立反相高效液相色谱法测定人血浆中紫杉醇的浓度。方法采用Dima kromasil C18柱;以地西泮为内标;以蛋白沉淀法提取血浆中的紫杉醇;以乙腈-水(53∶47)为流动相;流速为1.0 ml.min-1;检测波长为227 nm;进样量为20μl。结果血浆中紫杉醇线性范围为0.078~5.0 mg.L-1,相关系数r=0.999 9(n=8);平均提取回收率82.97%;平均加样回收率93.29%,日内、日间精密度RSD均小于10%。结论该法取样量少,操作简单,分离完全,结果可靠,回收率高,适用于临床紫杉醇的血浆中药物浓度监测和药代动力学研究。
Objective To determine the concentration of paclitaxel in human plasma by reverse phase high-performance liquid chromatography.Methods Dima kromasil-C18 was packed in the analytical column,diazepam used as the internal standard.The plasma samples were extracted by the method of protein precipitation-acetonitrile.The mobile phase consisted of acetonitrile:water(53∶ 47),with a flow rate of 1.0 ml·min-1,and the drug was detected at 227 nm by an ultraviolet detector.Results A better linearity was obtained in the range of 0.078~5.0 mg·L-1 for human plasma with a good correlation coefficient(r=0.999 9,n=8).The mean extract recovery was 82.97% and the mean recovery 93.29%.The detection limits were 38 μg·L-1,and the intraday and interday validations were less than 10%.Conclusion The improved method is sensitive,specific,simple,and suitable for the clinical drug monitoring of paclitaxel and pharmacokinetics study.
