摘要
目的建立测定多西紫杉醇脂质体药物含量及包封率的HPLC法。方法采用Diamonsil C18柱(250mm×4.6mm,5μm):流动相:甲醇-乙腈-水(体积比35:40:25);紫外检测波长:230nm。结果在本色谱条件下多西紫杉醇与辅料及溶荆峰分离良好,多西紫杉醇在1.0~50.0μg/mL范围内线性关系良好(r=0.9996,n=7),平均回收率为101.51%,RSD为1.96%。结论该方法准确可靠、简单快速,可用于多西紫杉醇脂质体含量及包封率的测定。
Objective To establish an HPLC method for the determination of content and entrapment efficiency of docetaxel in docetaxel liposomes. Methods The analysis was performed on a C18 column (250 mm×4.6 mm,5μm), The mobile phase consisted of methanol-acetonitrile-water (35: 40: 25) ,and the detection wavelength was 230 nm. Results The linear range for docetaxel was 1.0 - 50.0 μg/mL (r =0. 9996,n = 7). The average recovery was 101.51% and RSD was 1.96%. Conclusion This method is simple, accurate, sensitive for measurement of content and entrapment efficiency of docetaxel in the liposomes .
