摘要
目的建立测定多西紫杉醇脂质体药物含量及包封率的HPLC法。方法采用Diamonsil C18柱（250mm×4．6mm，5μm）：流动相：甲醇-乙腈-水（体积比35：40：25）；紫外检测波长：230nm。结果在本色谱条件下多西紫杉醇与辅料及溶荆峰分离良好，多西紫杉醇在1．0～50．0μg／mL范围内线性关系良好（r=0．9996，n=7），平均回收率为101．51％，RSD为1．96％。结论该方法准确可靠、简单快速，可用于多西紫杉醇脂质体含量及包封率的测定。
Objective To establish an HPLC method for the determination of content and entrapment efficiency of docetaxel in docetaxel liposomes. Methods The analysis was performed on a C18 column （250 mm×4.6 mm,5μm）, The mobile phase consisted of methanol-acetonitrile-water （35： 40： 25） ,and the detection wavelength was 230 nm. Results The linear range for docetaxel was 1.0 - 50.0 μg/mL （r =0. 9996,n = 7）. The average recovery was 101.51% and RSD was 1.96%. Conclusion This method is simple, accurate, sensitive for measurement of content and entrapment efficiency of docetaxel in the liposomes .
