摘要
目的:探讨B71单独或者与IL6联合在体外诱导T细胞活化的能力。方法:在混合淋巴细胞肿瘤细胞培养(MLTCs)后,测定淋巴细胞增殖指数和CTL杀伤活性。结果:B71+的B16细胞能够较有效地刺激淋巴细胞增殖、诱导特异CTL杀伤活性(与野生型B16细胞和模拟转染的B16细胞比较,P<0.01),当与IL6结合时效果更加显著(与各对照组比较P<0.01)。结论:B71基因在肿瘤细胞中表达可增强其免疫原性,在体外有效诱导T细胞活化;在此过程中IL6与B71分子存在协同效应。
Objective:To investigate the effects of B7 1 costimulatory molecule alone, or combined with IL 6, in inducing T lymphocytes activation in vitro.Methods:After splenocytes from non immunized C57BL/6 mice had been incubated with wild type, mock transfected, and murine B7 1 cDNA transfected B16 melanoma cells (B16 wt, B16 neo, and B16 mB7.1) respectively for six days, the lymphocytes were tested for both proliferation indices and cytotoxic activity against B16 wt cells.Results:Compared with B16 wt or B16 neo cells, B16 mB7.1 cells more effectively induced the proliferation of effector lymphocytes and the generation of specific lytic activity in vitro (P<0.01); The effects were more significant while combined with IL 6 (compared with other groups, P<0.01), though IL 6 alone could only induce effective proliferation, but not cytotoxicity. Conclusion: The costimulation signal of B7.1 is required for the activation and proliferation of T lymphocytes, the B7 1 expression in tumor cells could increase their immunogenicity and induced effectively T lymphocytes activation, and the cooperation of IL 6 and B7.1 costimulation is more effective.
