和解聚散方对单侧输尿管梗阻大鼠肾组织α-SMA表达的影响 被引量:6

摘要 目的:观察和解聚散方对单侧输尿管结扎(UUO)大鼠肾组织中α-平滑肌肌动蛋白表达的影响,初步探讨其抗肾间质纤维化的作用机制。方法:采用单侧输尿管结扎致肾间质纤维化大鼠模型,将雄性SD大鼠120只随机分为4组:假手术组,模型组,洛丁新组,和解聚散方组,每组30只,从造模后第2天开始灌胃,洛丁新组为1.0mg/kg/d,和解聚散方组为9.8g/kg/d,假手术组、模型组灌胃生理盐水3ml/只。分别于造模后7,14,21天取梗阻侧肾脏进行HE、Masson染色,观察肾组织病理结构变化;采用免疫组化方法检测肾组织α-平滑肌肌动蛋白(α-SMA)的表达。结果:模型组、洛丁新组、和解聚散方组中α-SMA在术后7天、14天、21天表达均明显高于假手术组,两个治疗组的表达水平明显低于模型组,和解聚散方组14天与洛丁新组14天组比较有显著性差异。结论:和解聚散方可以降低肾组织中α-SMA的表达,这可能是和解聚散方抗肾纤维化的作用机制之一。 Objective : To observe the effects of Hejiejusan decoction on the expression of α - SMA in kidney of UUO rats, and to discuss therapeutic function and mechanism of Hejiejusan decoction in renal interstitial fibrosis. Methods: The model rats were induced by ligating the right ureter. 120 male SD rats were randomly divided into four groups ( n = 30) : sham - surgery group, UUO group, Benazepril group and Hejiejusan decoction group. Gastric garage started from the day after surgery, benazepril 1.0mg/kg/d for Benazepril group, Hejiejusan decoction 9.8g/kg/d for Hejiejusan Decoction group, isotonic Na chloride 3ml per rat for sham - sur- gery group and UUO group. Rats were sacrificed on day 7, 14, 21 after UUO was initiated. Sections of the rehal tissue were stained with HE and Masson, which were used for histological and morphometric studies of the pathological change of the obstructed kidney. Immunohistochemical method was performed to examine the expression of α - SMA in the obstructed kidney. Results : Immunohistochemical staining showed that the level of - SMA of UUO group, Benazepril group and Hejiejusan Decoction group were significantly higher than that of sham - surgery group on day 7, 14, 21 after UUO, and the level of the two treatment groups was significantly lower than that of UUO group. Obvious difference existed between Hejiejusan decoction and Benazepril group after 14 days after UUO. Conclusion: Hejiejusan decoction exerts the anti -fibrosis effect in renal interstitial fibrosis, and it may be through decreasing the expressions of α - SMA in UUO rats.
