摘要
目的:运用网络药理学方法探寻保肾通络方防治DN的靶点和机制,为保肾通络方进一步的研究寻找方向和理论支撑。方法:通过BATMAN-TCM数据库筛选保肾通络方的活性成分及作用靶点;并通过Genecards数据库获取DN相关靶点;应用Cytoscape 3.7.0软件预测这些化合物对DN的作用靶点,构建药物-成分-靶点-疾病网络;应用R Studio软件进行基因本体(GO)分析,应用Cytoscape 3.7.0 clue GO插件进行京都基因与基因组百科全书(KEGG)富集分析,获取保肾通络方治疗DN的潜在作用通路。结果:保肾通络方经筛选后获得73个活性成分,58个可能参与治疗DN的靶点,主要参与营养水平、血压水平、血管形态和功能等方面的生物学过程,可能通过糖尿病并发症的晚期糖基化终末产物(AGEs)-糖基化终末产物受体(RAGE)信号通路、流体剪切应力与动脉粥样硬化、肿瘤坏死因子(TNF)信号通路、白介素-17(IL-17)信号通路、缺氧诱导因子-1(HIF-1)信号通路、肾素分泌等通路发挥其对DN的治疗作用。结论:保肾通络方防治DN的机制可能与其调节糖脂代谢、改善血流动力学,抑制炎性反应等有关。
Objective:To explore the possible targets and mechanism of using Baoshen Tongluo Recipe(BSTLD)in the treatment of diabetic nephropathy(DN)based on network pharmacology.Methods:Using BATMAN-TCM database to screen the active components and targets of BSTLD.Genecards database was used to obtain DN related targets.Cytoscape3.7.0 was used to predict the targets of these compounds for diabetic nephropathy,and a drug-component-target-disease network was constructed.GO enrichment analysis was performed using R Studio software.KEGG enrichment analysis was performed by Cytoscape3.7.0.Potential pathways of BSTLD in treating DN was obtained by clue GO plugin.Results:Seventy-three active components of BSTLD was screened and confirmed.Fifty-eight targets may participate in the treatment of DN,which were involved in biological process of the regulation of nutrition,blood pressure,and the morphology and function of blood vessels.BSTLD might play a role of the treatment of DN through AGEs-RAGE signaling pathway,fluid shear stress and atherosclerosis,TNF signaling pathway,IL-17 signaling pathway,HIF-1 signaling pathway,and renin secretion.Conclusion:BSTLD has a therapeutic effect on DN mainly through the mechanism of the regulation of glucose and lipid,improvement of hemodynamics,and inhibition of inflammatory response.
