摘要
目的:探讨组蛋白去乙酰化酶抑制剂Apicidin对Tca-8113细胞增殖及细胞凋亡的影响。方法:倒置显微镜、荧光显微镜观察Apicidin对Tca-8113细胞的影响;CCK-8法观察24h、48h、72h不同浓度Apicidin对Tca-8113细胞增殖的影响;DNA ladder检测细胞凋亡;流式细胞术检测凋亡率。结果:不同浓度的Apicidin对Tca-8113细胞均有一定的增殖抑制作用,且呈量效和时效关系;Apicidin能诱导Tca-8113细胞的凋亡且随着时间和剂量的延长越明显。结论:组蛋白去乙酰化酶抑制剂Apicidin能抑制人口腔鳞癌Tca-8113细胞增殖,诱导细胞凋亡。
Objective： To investigate the effect on Tca-8113 cells proliferation and apoptosis induced by histone deacetylase inhibitor Apicidin.Methods： Tca-8113 cells were stimulated with certain concentrations for 24h,48h,72h and were observed by light and fluorescence microscope.The influence on cell proliferation activity were determined by CCK-8 method.The process and rate of cell apoptosis were detected by DNA ladder and flow cytometry,respectively.Results： The inhibition on Tca-8113 cells proliferation was observerd under each concentration of the inhibitor and showed dual relations which include quantity and time.Apicidin can induce Tca-8113 cell apoptosis increasingly depending on time or dose.Conclusion： Histone deacetylase inhibitor Apicidin is able to significantly inhibit the proliferation and induce the apoptosis of Tca-8113 cells.
