温度对中国被毛孢液体培养的影响 被引量:2

Influence of Temperature on Liquid Culture of Hirsutella sinensis

摘要 通过改变温度,摇床培养中国被毛孢,根据时间变化,检测菌体干重,采用蒽酮-硫酸法检测菌体多糖含量,高碘酸钠比色法检测菌体甘露醇的含量,绘制了菌体液体生长曲线及菌体活性成分变化曲线,对中国被毛孢的液体生长规律进行了研究。结果表明:中国被毛孢的液体发酵周期为20 d;18℃培养16 d,菌体生物量达到最大,干重为0.789 mg/mL;菌体多糖含量最高,为4.73%;20℃培养16 d,菌体甘露醇含量最高,为3.54%。 To study liquid growth law of Hirsutella sinensis, Hirsutella sinensis was cultured with shake bed and temperature changing. According to the time change, dry weight of bacteria was detected, polysaccharide content in mycelium was detected by anthrone-sulfuric method, and mannitol content in bacteria was detected by sodium periodate colorimetric method. Meanwhile, the liquid growth curve of bacteria and change curves of bacteria active ingredient were drew. The results showed that the liquid fermentation period of Hirsutella sinensis was 20 d; when cultured at 18℃ for 16 d, the biomass and the polysaccharide content in bacteria all reached maximum, which were 0.789 mg/mL and 4.73%, respectively; when cultured at 20% for 16 d, the mannitol content in bacteria reached maximum which was 3.54%.
